Express custom monoclonal antibody production normally takes 8 weeks and is divided into 3 phases. We use ImmumEasy Mouse Adjuvant to save time with immunization, and use methylcellulose-based selection medium to save the time in screening clones. ImmunEasy Mouse Adjuvant is a new type of adjuvant specifically designed for inducing high antibody titers in mice. The adjuvant contains immunostimulatory CpG DNA short oligonucleotides that contain unmethylated cytosine-guanine dinucleotides within a certain base context. The mammalian immune system has evolved to recognize these sequences, which are found naturally in bacterial DNA, as a sign of infection. Exposure to CpG DNA results in very rapid and strong immune activation, and, when applied with an antigen, CpG DNA produces high titers of antigen-specific antibodies. The antibody titer induced by ImmunEasy Mouse Adjuvant is generally equal to or higher than that induced by Freund's complete adjuvant. In addition, high antibody titers are induced using up to 10-fold less antigen compared to some other adjuvants. Furthermore, boosting can be performed after 2 weeks instead of 4 weeks, saving both time and animal-housing costs. The immune response induced by CpG DNA is based on a natural reaction. Therefore, unlike many oil-based adjuvants, ImmunEasy Mouse Adjuvant does not induce symptoms of toxicity in immunized animals, even at doses much higher than those required for adjuvant activity. ImmunEasy Mouse Adjuvant can be used with most types of antigen, including small molecules, peptides, soluble proteins, virus-like particles, whole cell preparations, and polysaccharide conjugates. Since different CpG DNA sequences activate the immune systems of different animal species, ImmunEasy Mouse Adjuvant is specifically designed for immunization of mice.
The client has the option of halting the hybridoma production prior to the beginning of the next phase. We will not proceed to the next phase of the project without written (email) confirmation from the client. The amount of material needed for immunizations of 5 mice is 2mg to 3mg and an additional 2mg is needed for ELISA screening.

Hybridoma Technology
Phase I: A ready-to-use aqueous suspension of ImmunEasy Mouse Adjuvant and antigen are mixed, incubated for 5-10 minutes at room temperature, and then injected into 5 Balb/c mice for Immunization. Boosting is performed 2 weeks after the primary immunization.
Phase II: Fusion of spleen cells with myeloma cell line, plating the fusion product into six 96 well plates, and determination of those wells expressing antibodies to the antigen by ELISA assay. Culture fusion cells with methylcellulose-based selection medium containing HAT during a period of 9 days. All antibody-secreting colonies are isolated and transferred to a 24 well plate, expanded and a portion frozen. Antibody containing medium (3ml) of up to 12 ELISA-positive preclones* will be shipped to the client for evaluation in the client''s specific application.
Phase III: Cloning of positive (IgG secreting) wells, and take the clones culture in liquid medium containing HT in 1-2 days. A maximum of 5** of the positives selected by the client will be cloned by limiting dilution and isotyped. Clones will be expanded, and 10 vials of each selected clone will be frozen. Screen supernatants and positive wells, and clone them once. Medium (10ml) from each clone as well as the frozen vials can be shipped to the client upon request. Additional clones can be isolated and processed based on mutual benefits.
All resulting hybridomas, their antibodies, and reagents supplied are solely the property of the client. At the completion of the project, these items will be returned to the client.
